Nicotinic acid adenine dinucleotide phosphate (NAADP) regulates the T cell receptor induced Ca2+ signal and thereby modulates activation and differentiation of T cells. However, the role of different Ca2+ channels targeted by NAADP, specifically the type 1 ryanodine receptor (RYR1) and the two pore channels (TPC) 1 and 2, in T cell responses remain unclear. We will use mice with either complete or T cell restricted deficiencies of TPC1, TPC2 and RYR1, as well as of D-myo-inositol 1,4,5-trisphosphate (IP3) receptors, to study the function of these Ca2+ channels in T cells in vitro, and in infection and intestinal inflammation models in vivo.
