We hypothesize that soluble factors derived from adipose tissue under inflammation, such as ATP and ADO, differentially control dendritic cell (DC) migration. In order to test this hypothesis we will: (i) analyze purinergic signaling during the migration of murine DCs, by using microfluidic devices; (ii) evaluate Ca2+ signaling during the DC migration, with a focus in the ER, by combining live-cell imaging and Ca2+ sensitive fluorescent probes; (iii) analyze the effect of AN-derived from adipose tissue over DC migration in different levels of complexity, 2D within microfluidics, 3D in collagen matrix, and in vivo in mouse models.
